Sucrose Preference Test

Pleasure deficit is one of the frequent behavioral manifestations of depression, in which individuals are unable to experience pleasure from rewards or other pleasurable things. In animal behavior experiments, sucrose preference test is commonly used to test for pleasure deficit symptoms.

Experimental Principle

Sucrose preference test exploits the rodent's innate strong preference for sweetness. In sucrose solution and plain water, rodents will selectively drink the sweet sucrose solutions. However, when rodents are in a depressed model, they lose interest in sweetness. Therefore, detecting the preference of animals for sucrose solutions can be a useful tool to assess the symptoms of pleasure deficit and the degree of depression in animals.

Experimental Flow

The entire sucrose preference test consists of three phases: adaptation, baseline measurement and testing. Here, only a brief description of the process of the sucrose preference test is introduced. We can design and modify the test protocol to suit the specific demands of our customers' researches, such as incorporating animal modeling, dosing times and other factors.

  • Adaptation period (Days 1 to 4)

Two drinking units containing 1% sucrose solution and plain water were placed in corresponding positions in the experimental test chamber and changed daily to avoid location preference of the animals.

  • Baseline measurement

The experimental animals were placed in test chambers containing sucrose water and plain water, and the consumption of both solutions by the animals was recorded after 12 hours. After 24h, the positions of the two solutions were changed and the consumption of the solutions was recorded again.

The two sets of data from the baseline test were recorded for subsequent data comparison and analysis.

  • Testing period

After 24h of water fasting, the sucrose water preference index was measured. The consumption of both solutions by the animals after 12h was recorded.

Sucrose preference (%) = sucrose water consumption / (sucrose water consumption + drinking water consumption) × 100%.
